چکیده

Protoplasts of Convolvulus arvensis L. tissue culture regenerated a wall-like structure within 3 days in culture. Although unusually electron dense and atypically amorphous in the electron microscope, this structure could be digested with Myrothecium cellulase but was resistant to protease, a Rohm and Haas pectinase, and a beta-1, 3-exoglucanase just like the original wall. A cytochemical test for callose was negative. Wall regeneration required a readily metabolized external carbon source and was not inhibited by a high concentration of cycloheximide, puromycin, or actinomycin D. Protoplast budding was correlated with the wall regeneration, and the latter was related quantitatively to the sucrose concentration in the medium. Although a concentration of 1 mum 2,4-dichlorophenoxy acetic acid is used normally for both general culture of the tissue and for wall regeneration, concentrations of 0 and 0.1 mm, which are highly deleterious to growth, have no appreciable effect on the incidence of the wall-like structure regenerated around protoplasts. The ability of protoplasts to undergo cell wall regeneration was decreased when they were cultured in the presence of proteolytic enzymes.

SHOOT REGENERATION FROM SAFFRON PROTOPLASTS IMMOBILIZED IN Ca-ALGINATE BEADS

Saffron (Crocus sativus L.) protoplasts were isolated from the cells of a suspension culture or calli with a solution of Cellulase, Pectinase and Hemicellulase and embedded in Ca-alginate beads. They were cultured with or without nurse cells in MS medium supplemented with 2,4-D and 6-benzylaminopurine at 25°C. After several changes of medium, cell-clusters appeared on the surface of the Ca-algi...

A simplified Protocol to Induce Callogenesis in Protoplasts of Date Palm (Phoenix dactylifera L.) Cultivars

Background: In Algeria, date palm is currently confronted to the Bayoud disease. Biotechnological tools such as protoplasts fusion can appear as an alternative to ensure rapid multiplication and improvement of this species. Objectives: Callogenesis induction in protoplasts isolated from embryogenic callus of three date palm cultivars. Materials and Methods: Some factors influencing the isolatio...

SHOOT REGENERATION FROM PROTOPLASTS OF PENNISETUM AMERICANUM L (MIL CEREAL)

A finely dispersed cell suspension was obtained from embryogenic calli of two genotypes of pearl millet (Pennisetum americanum L. Mil cereal). Protoplasts isolated from both cell suspensions divided and produced globular embryoids and calli. On an agar medium, the embryoids were induced to form albino shoots or albino plantlet- like structures

Plasma membrane ultrastructure during plant protoplast plasmolysis, isolation and wall regeneration: a freeze-fracture study.

The freeze-fracture morphology of the plasma membrane of cells and isolated protoplasts of plant callus suspensions has been investigated. Plasmolysis of suspension cells leads to the formation of 2 types of hexagonal arrays of intramembrane particles situated on the inner fracture face (PF). These arrays are interpreted as proteins that have 'crystallized' in the plane of the membrane as the a...

PLANT REGENERATION FROM MESOPHYLL PROTOPLASTS OF POTATO (SOLANUM TUBEROSUM L.) CULTIVAR DELAWARE USING SILVER THIOSULFATE (STS)

The use of in vitro potato shoot cultures for protoplast isolation is desirable for consistent protoplast qualities. Ethylene build-up in the culture vessels causes problems for leaf growth (e.g. decreasing leaf surface, dry weight and chlorophyll) in shoot culture. Fifty to one hundred µM Silver thiosulfate (STS) produced a larger leaf area as a source of plant material for protoplast isolatio...
